NaCl对木枣试管苗继代培养的影响研究

摘要:以木枣(Zizyphus jujube cv. Muzao)试管苗带芽茎段为外植体,在试管苗继代培养基中加入不同浓度的NaCl(1、5、10、20 mg/mL),考察其对木枣试管苗外植体高度和粗度、鲜重和干重、繁殖系数、叶绿素含量等指标的影响。结果表明,木枣试管苗外植体在盐胁迫下表现出生长缓慢、分生能力减弱、丛生芽簇生、叶片颜色变浅、茎段变粗等现象,木枣外植体的繁殖系数、平均高度、鲜重和干重均随着NaCl浓度的升高逐渐降低,叶绿素含量随NaCl浓度的升高呈先升高后降低的变化趋势,NaCl浓度为1 mg/mL时叶绿素含量最高。

Abstract: Taking stems with buds of Zizyphus jujube cv. Muzao as explants, NaCl was added in the subculture medium at different concentrations to investigate its effects on the height and diameter, fresh and dry weight, propagation coefficient and chlorophyll content of explants. The results showed that under salt stress, the explants had slower growth rate, weaker differentiation ability, tufty multiple shoots, shallower leaf color and thicker stems. With the increase of NaCl concentration, the propagation coefficient, average height, fresh and dry weight decreased; chlorophyll content increased firstly and then decreased, was the highest when NaCl concentration was 1 mg/mL.

枣树(Zizyphus jujuba Mill.)为中国特有树种,有“铁杆庄稼”的美誉,我国有700多个枣树品种,在陕西分布有79个枣树品种,其中木枣(Z. jujube cv. Muzao)为陕北枣区的主栽品种。对陕北地区枣树的研究主要集中在高产栽培、果实保鲜、天然产物提取及应用、生物活性成分分析、组培育苗、芽变品种选育等方面[1-7],而对该地区盐害对枣树的影响方面的研究很少。盐胁迫会造成植物发育迟缓,抑制植物组织和器官的生长和分化,最终导致植物产品产量和质量的下降[8]。开展枣树耐盐性研究,进而了解枣树抵抗盐害胁迫的生理机制,对于枣树耐盐品种选育和高产栽培具有重要的理论和实践意义。因此,本研究对NaCl胁迫对木枣试管苗继代培养的影响进行了分析,以期为木枣组培快繁和试管苗生理生态研究提供依据。

1 材料与方法

1.1 材料
